I. Oxidative stress
지금으로부터 약 100여 년 전, 생체 내의 물질대사 속도(metabolic rate)가 빠를수록 수명이 더 짧다는 것이 알려지면서 물질대사 속도가 궁극적으로 기대 수명을 결정한다는 ‘the rate-of-living hypothesis`가 대두되었다. 그리고 1950년대 중반, 세포 내에서 발생한 활성 산소 종(Reactive Oxygen Species, ROS)으로 인하여 축적된 damage가 노화를 일으킨다는 ’free-radical theory of aging`이 제안되었다. 특히, superoxide anion을 제거하는 super- oxide dismutase(SOD)의 발견은 이러한 물질대사와 노화 간의 연관성을 설명하는 중요한 단초를 제공하였다. McCord, J. M. &Fridovich, I. Superoxide dismutase. An enzymatic function for erythrocuperin (hemocuperin). J. Biol. Chem. 244, 6049 6055 (1969).
또한, 세포 내의 대부분의 에너지를 생산하는 mitochondria가 세포 내 산소의 상당량을 소모한다는 사실이 밝혀지면서 현재는 ‘free-radical theory of aging’과 ‘the rate-of-living hypothesis`를 동일한 의미로 받아들이고 있다. 즉, 생체 내 물질 대사 속도가 빠를수록 ROS의 생성량이 증가하여 수명이 감소한다는 것이다.
