3. 기본 배경
■ GABAA receptor → Ligand-gated channel
● GABAA receptor의 구조
The receptor is a pentameric transmembrane receptor that consists of five subunits arranged around a central pore. Each subunit comprises four transmembrane domains with both the N- and C-terminus located extracellularly. The receptor sits in the membrane of its neuron, usually localized at a synapse, postsynaptically. However, some isoforms may be found extrasynaptically.
● GABAA receptor의 특징
The GABAA receptor is an ionotropic receptor and ligand-gated ion channel. Its endogenous ligand is γ-aminobutyric acid (GABA), a major neurotransmitter in the central nervous system. Although GABA ends up producing an inhibitory effect on neurotransmitter release, GABA is not an inhibitory neurotransmitter itself as it stimulates the GABA receptor; therefore, directly it is a stimulatory neurotransmitter. Upon activation, the GABAA receptor selectively conducts Cl- through its pore, resulting in hyperpolarization of the neuron. This causes an inhibitory effect on neurotransmission by diminishing the chance of a successful action potential occurring.
