1. Explain and compare thin layer chromatography with column chromatography.
Thin Layer Chromatography (TLC) is used to separate the pure individual compounds from a mixture. TLC is one of a quick and inexpensive chromatographic technique that is most widely used to monitor the progress of a reaction by comparing the Rf of a compound with Rf of a known compound.
<Discussion>
In this investigation, many different ratios of eluent systems (mixture of ethyl acetate: hexane) were used to separate and assist the compounds: aniline (A), benzoic acid (B) and triphenylphosphine (C) on the silica gel by capillary action. The main objective of this study was to find appropriate eluent composition that can determine Rf values within 0.3-0.4 for each compound by using TLC. Therefore, our group started with 1:7 (EA: hexane) and found that Rf values for compounds A, B and C were: 0.25, 0.469 and 0.781, respectively.
