<Experimental Background>
Protein purification is a technique used to isolate a single protein type that is usually extracted from a sample of biological tissue or complex mixture such as a cell lysate. Many times, single specific protein interest is caught in a matrix of other protein or even non-protein parts and hence protein purification must be done to separate the single desired proteins from all other mixtures.
<Results/Discussion>
AKTA FPLC purification of His-tagged protein was performed and following curves were attained after applying the samples through immobilized Ni-columns. 10mM binding buffer & 500mM elution buffer (with imidazole) were used.
