The concentration of isolated plasmid DNA is measured by spectrophotometric analysis using Thermo Scientific NanoDropTM spectrophotometer. Here, the concentration of mini-prep plasmid DNA is 110.9ng/μL. In addition, the recovery and purity of isolated plasmid DNA can also be determined by spectrophotometric analysis.

3. Describe the unit of restriction enzyme activity.
Enzymes are quantitated in “unit” and so by knowing the unit of restriction enzyme (RE), the amount of RE required to digest certain concentration of DNA can be determined. Hence, 1 unit indicates the amount of RE needed to digest 1μg of λ DNA in a reaction volume of 50μL in 1 hour at optimal temperature of a particular enzyme.
