1. Chymotrypsine is a well-known enzyme whose active site (Ser-His-Asp triad) is elucidated. Draw chymotrypsin using a protein visualization tool while clearly presenting the chemical motifs of the triad in chemical structures. See an example of carbonic anhydrase below; it has a histidine triad coordinated with Zn ion.
Chemical motifs of the triad in chemical structures
The chymotrypisin is shaped like a sphere and consists of three polypeptide chains, which are connected by a disulfide bond. It is composed of a single polypeptide called chymotrypsinogen, which is activated by dividing the polypeptide into three chains with a protein breakdown enzyme.
The side chain of serin 195 is combined by hydrogen bond, with the imidazole ring of the histidine 57. And the NH group of the imidazole ring is combined by hydrogen bond with carboxyl group.
Like this, the group of residues is called catalytic triad.
