5. Method
<Making agar culture medium>
① Suspend 20.0g of the TSA powder in 500ml of purified water in 3 Erlenmeyer flasks each.
② Mix thoroughly with Magnetic bar and Stirrers. and Heat with frequent agitation and boil for few minutes to completely dissolve the powder.
③ Autoclave at 121˚C for 15 minutes.
④ Cool enough to about 50˚C.
⑤ Using a pipette, place liquid agar in each 2 test tube and petri dish. and Close the lids.
* When create a slanted agar, test tube should be maintained in the proper angle.
* When pour the agar, It should be done within the clean bench.
* Be careful not to make bubbles on center of the agar.
* If the amount of agar is too small, bacteria can not grow well because of malnutrition.
* If after one hour of hardening of the agar, plate upside down to dry because of moisture.
<Inoculating E. coli>
① Sterilize the loop by holding the wire in the flame until it is red-hot.
② While holding the sterile loop and the bacterial culture, remove the cap.
