Flow Cytometry(유세포 분석기)는 유핵 상태의 입자나 세포가 Flow 상태에서 Sensing point(검출영역)을 지날 때 핵내의 DNA양의 분석이나 세포의 크기, 표면항원, 내부조성 등의 차이 또는 변화를 분석할 수 있다. Flow Cytometry 는 Fluidic system, Optical system, electronic system 으로 구성되며, Flow Cytometry의 장점은 짧은 시간 내 수많은 입자나 세포에 대한 양적, 질적인 측정을 하여 검체 내 입자나 세포 하나하나의 고유한 특징을 알아 낼 수 있다는 점이다. 특히 모든 검출은 single-cell level로 이루어지며 각각의 세포마다 여러 형광물질을 동시에 사용하여 검색하므로 다양한 물리적, 화학적 특성을 한 검체에서 측정하여 객관적인 계량화가 가능하다는 장점이 있다. 임상적으로는 림프구 특성 분석, Lymphocyte leukemia와 lymphoma의 성상규명, DNA content 분석, 혈소판 연구 및 각종 면역질환의 진단 및 병인 연구에 널리 사용되며 면역학, 세포생물학 및 발생학 분야 등에서 매우 광범위하게 이용되고 있다. 진단검사학적으로는 혈액학적인 cell counter로 그 원리가 우리에게 익숙하지만 그 적용 범위는 많은 차이를 보인다. 연구소에서 많이 사용되어지던 Flow Cytometry가 임상적 검사에 사용되어진 것은 몇 년이 안 되지만 현재 사용되고 있는 검사 종목의 임상적 중요성과 Flow Cytometry에 대한 진단검사학적 응용의 무한한 가능성에 대해 논하고자 한다.Ⅱ. 본 론
1. Flow Cytometry 의 발전 역사
1930년 스웨덴 Karolinska Institute의 Caspersson이 세포내 핵산과 단백질 양을 측정하기 위해 microspectrophotometer를 개발한 것이 가장 시초이다. 1947년 Northwestern대학(미국)의 Gucker등에 의해 공기를 빠른 속도로 주입하면서 검체를 실려 보내고 입자가 통과하면 빛이 산란되어 photodetector에서 전기 신호를 검출해낼 수 있는 최초의 Flow cytometer가 개발 되었다.
